Once mature and in place, melanocytes produce melanin, the pigment responsible for skin colour, which is exported to the surrounding keratinocytes (each melanocyte is connected to roughly 40 keratinocytes) in grain-like structures known as melanosomes. So, what’s the major function of melanin, you ask? Melanocytes reside in the basal level of the epidermis at the base of hair follicles and possess many long dendrites that allow intercellular transport of pigment containing melanosomes from the cell body, the site of synthesis, to surrounding keratinocytes (for review see Jackson 1994). Melanophores (or melanocytes in humans) contain melanin and are the most common type of chromatophores, responsible for the black, brown, and brownish red coloration of vertebrates. Melanophores contain thousands of melanosomes that vary in size, shape, and composition depending on the type of melanin they contain. Melanosomes are specialized intracellular organelles that produce and store melanin pigments in melanocytes, which are present in several mammalian tissues and organs, including the skin, hair, and eyes. Melanosomes form and mature stepwise (stages I–IV) in melanocytes and then are transported toward the plasma membrane along the cytoskeleton.

Melanosomes are unique organelles in melanocytes that produce melanin, the pigment for skin, hair, and eye color. Tyrosinase is the essential and rate-limiting enzyme for melanin production, that strictly requires neutral pH for activity. pH maintenance is a result of the combinational function of multiple ion transport proteins. Melanocytes produce specific organelles, termed melanosomes, in which melanin pigment is synthesized and deposited. In the skin, melanosomes are transferred from melanocytes to neighboring keratinocytes in order to form perinuclear melanin caps (Hearing, 2005). Melanocytes play a major role in protecting the skin from ultraviolet radiation through the production of melanosomes that are transferred to keratinocytes to protect their nuclei.
